It can be done, but the system would perceive it as a second multihosted domain. Since you get five free multihosted domains this would use two up.
There is some other threads were we talk about improving upon the mutihosted feature and we are working on that as well.

So let me make sure I have this right. If I wanted to have a subdomain of say, http://website.daisyhead.org on my domain daisyhead.org, which for this sake we'll say is a multi-hosted domain, that would count as being a 2nd mult-hosted domain?

yes. If daiseyhead.org is already multihosted on another domain then website.daisyhead.org would be a second multihosted domain.
